LabChip® GXII Touch毛细管电泳系统采用一种独立进样针的微流体芯片来快速检测24、96或384孔微孔板中的蛋白样本。配套相应的试剂盒,可自动染色、脱色、电泳分离和分析蛋白样本。LabChip仪器的光学组件检测到激光诱导荧光信号后,易于使用的系统软件可利用ladder和marker标记物自动校准并分析数据,为用户提供蛋白浓度、片段大小以及纯度分析。

 

ProteinExactTM HR用于6.5-250kD蛋白检测,尤其适合滴度检测,目前也有使用该款芯片进行AAV的滴度和检测重复性的开发。

Q:使用ProteinExact HR检测AAV的原理。

A:AAV衣壳由VP1、VP2和VP3三种亚单位组成,共60拷贝,比例为1:1:10(VP1:vp2:vp3),通过测定三种衣壳蛋白浓度的比例来识别AAV。

 

Q:实验时校准失败的原因是什么?

A:可能是标准面积测量值超出测定验证范围,或者是染料混合比与标准不同,也有可能是芯片本身质量问题。 

 

Q:出现实验失败问题怎么解决?

A:根据用户手册进行标准实验;重新准备芯片确保试剂新鲜。
